Instead of the single cell dispersion of vital dye-excluding cells which one obtains from normal liver, a mixture of cell clumps, few single cells, and largely undissociated tissue fragments is obtained. Attempts to mechanically disperse the perfused tissue into a single cell suspension lead to loss of cell viability. Because of these prob­ lems, it is often not possible to prepare oval cells and parenchymal cells from the same liver. Therefore, the perfused CDE liver tissue is next subjected to a second enzymatic digestion designed for isolation of either oval cells or parenchymal cells.
